您好:
感謝您使用本單位的微生物菌種,我們有可用于科研、教學(xué)、生產(chǎn)的菌種約2500株,基本覆蓋了環(huán)境、工業(yè)、農(nóng)業(yè)、分析、食、藥用真菌行業(yè)各類生產(chǎn)和科研用微生物,其中許多優(yōu)良菌株的生產(chǎn)性能具有較高水平。我們對(duì)所提供菌種作如下說(shuō)明:
菌種簡(jiǎn)介:
1、菌種名稱:ATCC 9027 銅綠假單胞菌(綠膿桿菌)
2、規(guī)格:斜面□ 凍干管□
二、保藏條件:凍干菌種應(yīng)在2-8℃保存;斜面菌種放15-20度保存,15-20天轉(zhuǎn)接一次。
三、培養(yǎng)條件:
1. 培養(yǎng)基:營(yíng)養(yǎng)瓊脂培養(yǎng)基(NA)
胰蛋白棟大豆肉湯培養(yǎng)基(TSA)
LB培養(yǎng)基 以上培養(yǎng)基三選一即可
2. 培養(yǎng)溫度: 37℃
3. 培養(yǎng)時(shí)間:18- 24h
四、真空凍干菌種的復(fù)蘇培養(yǎng):
1、安瓿管開(kāi)封:用浸過(guò)75%酒精的脫脂棉擦凈安瓿管,用火焰加熱其頂端,滴少量無(wú)菌水至加熱頂端使之破裂,用銼刀或鑷子敲下已破裂的安瓿管頂端。
2、菌種恢復(fù)培養(yǎng):用無(wú)菌吸管吸取 0.1-0.2毫升的培養(yǎng)液或無(wú)菌水,滴入安瓿管內(nèi)輕輕振蕩,使凍干菌體溶解呈懸浮狀,吸取全部菌體懸浮液,移植于1-2支建議的斜面培養(yǎng)基上,并在建議的溫度下培養(yǎng)。
五、注意事項(xiàng):
1、微生物菌種應(yīng)保藏于低溫、清潔和干燥的地方,室溫放置時(shí)間過(guò)長(zhǎng)會(huì)導(dǎo)致菌種衰退;
2、菌種操作應(yīng)在無(wú)菌條件下進(jìn)行,防止雜菌污染;
3、凍干菌復(fù)蘇后,應(yīng)根據(jù)菌種狀況及時(shí)轉(zhuǎn)接傳代
